GlowBioTM Caspase 3/7 substrate is a caged bioluminescent probe based on the DEVD peptide. The amount of light produced as the result of specific cleavage of the DEVD sequence is proportional to the activity of Caspase3/7. GlowBioTM is superior to the commercially available DEVD-aminoluciferin substrate (VivoGlowTM, Promega) for imaging of caspase 3/7 activity and is based on novel biocompatible ligation reaction6. It produces over 5 fold higher signal to background ratios when directly compared to VivoGlowTM in mice treated with d-GalN/LPS, a commonly used liver inflammation model6. The probe could also be successfully for sensitive imaging and quantification of physiological levels of Caspase 3/7 activities in mice treated with chemotherapy.
